The church was built between 1538 and 1545 by Antonio da Sangallo the Younger. The elevation was completed by Ottaviano Mascherino during the pontificate of Sixtus V, following Sangallo's design. "Besides many interesting designs for villas, other notable late works include the church of Santo Spirito in Sassia, noted mainly for its façade, which is a forerunner of later Counter-Reformation solutions.
